Cell-to-Cell Measles Virus Spread between Human Neurons Is Dependent on Hemagglutinin and Hyperfusogenic Fusion Protein

Measles virus (MV) usually causes acute infection but in rare cases persists in the brain, resulting in subacute sclerosing panencephalitis (SSPE). Since human neurons, an important target affected in the disease, do not express the known MV receptors (signaling lymphocyte activation molecule [SLAM] and nectin 4), how MV infects neurons and spreads between them is unknown. Recent studies have shown that many virus strains isolated from SSPE patients possess substitutions in the extracellular domain of the fusion (F) protein which confer enhanced fusion activity. Hyperfusogenic viruses with such mutations, unlike the wild-type MV, can induce cell-cell fusion even in SLAM- and nectin 4-negative cells and spread efficiently in human primary neurons and the brains of animal models. We show here that a hyperfusogenic mutant MV, IC323-F(T461I)-EGFP (IC323 with a fusion-enhancing T461I substitution in the F protein and expressing enhanced green fluorescent protein), but not the wild-type MV, spreads in differentiated NT2 cells, a widely used human neuron model. It has been the largest market for Apple iPhones since 2016. Since the escalation of tariffs, though, China has stopped buying soybeans and lobsters, and Apple warned it would miss its expected Christmas holiday sales figures because of trade tensions. In addition to the 25% tariffs below, Beijing also added 20% tariffs on 1,078 US products, 10% tariffs on 974 US products, and 5% tariffs on 595 US products (all links in Chinese). Christopher Slutman, a Marine and New York City firefighter, was taken to New York today, two weeks after the 43-year-old was killed while serving in Afghanistan. Slutman, who is survived by his wife and three daughters, was one of the soldiers killed by a roadside bomb on April 8 during a convoy near the Bagram Airfield in Kabul. Slutman had a residence in Yonkers but his family lived in Delaware, and today a procession took him from Dover Air Force Base to Joseph Lucchese Funeral Home in the Bronx. Firefighters honored him as his remains are driven over the Delaware Memorial Bridge, up the New Jersey Turnpike and over the George Washington Bridge into New York. The body of FDNY member and U.S. Marine Christopher Slutman was transported from Dover Air Force Base in Delaware to New York City Monday. For many decades, so-called crate engines—most often V-8s—have been the basis for adding more excitement or a life extension to old cars and trucks, fresh out of a shipping crate and with a minimum of fuss.  Electric GT, of Chatsworth, California, is in the process of doing a “TLC build” electric conversion of a 1970 Toyota FJ40 Land Cruiser. This conversion, which is nearly complete, also happens to be a testbed for an all-in-one modular “crate” system.  Eric Hutchinson and Brock WInberg are partners in Electric GT, which started a few years ago with a project to replace a Ferrari 308’s V-8 with an all-electric powertrain—creating one very quiet (and quicker) version of the car made famous on Magnum P.I.. The idea, at its core, Hutchinson explained to Green Car Reports, is to create a modular crate engine kit that can be assembled according to a number of customer choices, then sent to a builder, where it can reduce the amount of labor and setup time.
